the nucleotide excision repair create removal. this removal creates a single-strand gap in the DNA, which is filled in by DNA polymerase using the undamaged strand as a template and thereby restoring the original nucleotide sequence.
Rad gene is involved in nucleotide excision repair. His gene was amplified by PCR and inserted into the Rad gene. In the case of eukaryotes, knockdown is mainly used rather than knockout because intron exists, but yeast can eliminate the gene itself because there is no intron.
The rad gene knockout yeast have his gene by homologous recombination
method. Therefore, it will grow well on SD-His medium. Only such colonies were selected and the gene was obtained using a 10 min prep. After confirming by PCR, spotting is conducted.
In this study, we performed an experiment to identify the phenotype by knocking out the rad gene of Yeast. The phenotype was confirmed by spotting in SD4 +, SD4 + UV50J, SD4 + UV100J and SD4 + NQO 10ng / ml medium.
