    실험제목(title): Transformation
    · 실험날짜(date):
    · 실험목적(purpose): plasmid DNA를 competent cell에 도입하여 colony를 관찰함으로써 transformation과정을 이해한다.
    · 실험 준비물(material-reagent, instrument)
    피펫, LB agar 배지, spreader, DH5 alpha, 항온수조, 얼음수조, e-tube, dilution한 DNA

    · 실험 방법(method)
    ① DH5 alpha를 얼음이 담긴 수조에 두고, e-tube를 얼음에 두어 차갑게 유지한다.
    ② 20μl의 DH5 alpha를 e-tube에 넣는다.
    ③ DNA 2μl을 DH5 alpha에 넣는다. (최대한 competent cell이 충격을 받지 않도록 주의한다.)
    ④ 얼음에서 30분간 incubation한다.
    ⑤ 42℃에서 90sec간 incubation한다.
    ⑥ 얼음에서 5분간 incubation한다.
    ⑦ 37℃에서 데운 항생제가 들어있는 LB agar 배지에 competent cell 20μl을 넣고 spreading한다.
    (control group)
    ⑧ 37℃에서 데운 항생제가 들어있는 LB agar 배지에 transformation한 dna를 20μl 넣고 spreading한다.
    ⑨ 37℃에서 overnight으로 배양한다.

    · 실험 고찰(discussion): 실험에 대한 고찰도 추가하기
    competent cell은 온도와 충격에 민감하므로 실험 시 주의해서 다뤄야한다. 또한 배지에 spreading시 배지가 파이지 않도록 조심해서 다뤄야 한다.
