    본문내용

    Western blotting (immunoblotting)은 샘플 내 특정 단백질의 존재 여부, 양, 크기를 측정하는 실험이다. 단백질이 소량이어도 검출 가능하고, 다양한 종류의 단백질이 혼합된 시료를 검사할 때도 항체의 기질 특이성 덕분에 원하는 단백질만 검출할 수 있다는 장점이 있다. 전기영동을 통해 젤 내에서 단백질을 분리하여 이를 nitrocellulose 또는 PVDF 막으로 복사하듯 전달하고, 타겟 단백질에 특이적인 항체로 검출하는 방식으로 진행된다.
    이 방법은 크게 5개의 순서로 이루어져 있다. 첫째로, SDS-PAGE를 시행한다. 이는 SDS를 이용하여 단백질을 (-)전하를 띄도록 변성시켜 선형으로 만들고 전기영동을 통해 크기별로 분리하는 과정이다. 둘째, 분리된 단백질을 membrane에 전이한다. gel 위에 membrane을 얹고 gel쪽을 (-), membrane쪽을 (+)로 하여 전기장을 걸어주면 단백질이 (-) 전하를 띠므로 membrane쪽으로 이동한다. 셋째, blocking solution을 첨가하여 1차 항체와 membrane의 비특이적 결합을 방지한다. Blocking solution으로는 skim milk나 BSA를 사용한다. 넷째, 타겟 단백질과 결합하는 1차 항체를 투여한다. 다섯째, 결합하지 않은 항체를 씻어낸 후 2차 항체를 투여하여 밴드를 확인한다. 2차 항체는 1차 항체의 Fc 부분과 결합하고 발색효소와 컨쥬게이션 되어 있으므로 발색반응을 통해 밴드를 시각화할 수 있다. 2차 항체와 컨쥬게이션되는 발색효소 중 하나인 horse radish peroxidase(HRP)는 luminol과 같은 substrate를 산화시킨다. 산화된 luminol은 excited state으로부터 ground state로 전이하면서 빛에너지를 방출하므로 이를 검출한다.
