초록

Rumex japonicus Houttuyn is known for its antiinflammatoryand anti-microbial effects. In this study, wedetermined the effects of ethanol extract of Rumex japonicusHouttuyn (RJHE) on anti-obesity activities in 3T3-L1 preadipocytes.
In vitro experiments to prevent obesity and improveanti-obesity effects, research using 3T3-L1 preadipocytes isessential, it also distinctly represents morphologic and molecularchanges. Our results showed that RJHE decreased thedifferentiation of 3T3-L1 preadipocytesusing oil red O stainingthrough decreasing the lipid droplet and had no cytotoxicityin 3T3-L1 preadipocytes using WST-1 assay (100 μg/mLand 200 μg/mL). Also, the concentrations of triglycerides(TG)were gradually reduced in 3T3-L1 preadipocytes cells withRJHE (N: 100%, 100 μg/mL: 64.5%, 200 μg/mL: 38.1%) usingELISA assay. And RJHE suppressed the Expression of PPARγ,C/EBPα and Acetyl-CoA Carboxylase (ACC) in a dose-dependentmanner by Western blot analysis. Taken together, ourresults demonstrate that RJHE indicates anti-obesity effects invitro situation and has potential uses in chemotherapeutic drugs.
