I.Restriction enzymes
1)Definition
Restriction enzyme (or Restriction endonuclease) is an enzyme that cuts DA at or near specific recognition nucleotide sequences known as restriction site,

Restriction enzymes are traditionally classified into three type on the basis of subunit composition, cleavage position, sequence specificity and cofactor requirement. However, amino acid sequencing has uncovered extraordinary variety among restriction enzymes, there are many more than four different types.
- Type I: Type I cuts DNA at random locations as far as 1000 or more base-pairs from the recognition site. Enzymes possess three subunits for restriction, methylation, and sequence recognition. The cofactors S-Adenosyl methionine, ATP and MG2+ are required for their fully activity.
- Type II: Type II enzymes are predominantly used in biotechnology. Type II cut DNA at defined position close to or within their recognition sequences without the need for ATP.
