Title: Isolation, Microscopy, and Measurement of Chloroplast
Introduction:
Chloroplast is a cellular organelle that is responsible for photosynthesis, a process in which light energy is converted to chemical energy and stored in the form of sugar and other organic compounds. A chloroplast can be divided to two parts: grana and stroma. Grana consists of many coin-shaped structures called thylakoids, which contain chlorophylls, green pigments responsible for absorbing light during photosynthesis. Chlorophylls are what give plant leaves their green color. Stroma is the matrix between layers of grana and the outer layer of the chloroplast. The Calvin cycle takes place there, which is a process in which CO2 is converted to sugar using products from light reaction, which takes place at thylakoids. This process is called the dark reaction.
Cell homogenization is a process whereby a cell is fractionized and brought to a state such that all parts of the sample are equal in composition; that is, it is mixed so well that removing some of the sample does not alter the overall make-up of the remaining sample.
