초록

Blechnum orientale, also known as the centipede fern, is an important medicinal fern in Malaysia, India, and Chinese. The present study was designed to evaluate the antioxidant, anti-melanogenesis activities of an 80% aqueous methanolic extract of Blechnum orientale rhizome(BOR) and leaf (BOL). The antioxidant assays were perforemed using 2,2-diphenyl-1-picrylhydrazyl(DPPH) and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt(ABTS). The DPPH and ABTS radical scavenging activities of BOR extract were increased in a dose-dependent manner. Both BOR and BOL extracts did not affect human keratinocytes, HaCaT cell viability. Anti-melanogenesis activity was evaluated by inhibiting the mushroom tyrosinase enzymes, as this causes melanin content in murine melanoma, B16F10 cell line. The BOR extract significantly reduced tyrosinase activity and the cellular melanin synthesis in B16F10 cells without cytotoxic effects. Additionally, The BOR extract also significantly decreased the protein level of melanogenesis-related genes such as microphthalmia-associated transcription factor(MITF), tyrosinase-related protein(TRP)-1, and tyrosinase-related protein(TRP)-2. Collectively, these results indicated that cosmetics containing extract of B. orientale were safe and this plant extract could be used as a functional cosmetic material.
