1. Transfection

a. Transfection : `infection by transformation`

The introduction of a foreign gene into mammalian cells.
Transfection은 virus vector나 다른 transfer vector를 통해 foreign gene을 eukaryotic cell에 introduction 시키는 것을 말한다. transfection과 transformation 의 차이는 transfection은 주로 mammalian cell을 target으로 한다는 것이다.
transfected cell은 foreign DNA의 introduction으로 인해 그 properties가 변하게 된다.

b. Methods

foreign DNA를 eukaryotic cell에 introduction하는 방법에는 어떤 carrier를 쓰느냐 에 따라 여러 가지가 존재한다.
- calcium phosphate : phosphate ion을 포함하고 있는 HeBS(HEPES-bufferd saline solution)에 foreign DNA가 포함된 calcium chloride solution을 혼합하면 calcium ion과 phosphate가 반응하여 DNA와 함께 precipitate를 형성한다. 이 suspension을 cell에 첨가하면, cell이 precipitate를 uptake하게 된다.
