Photosynthesis에 관여하는 protein은 reaction center(RC)를 비롯하여 light harvesting complex(LHC), photosystemⅠ(PSⅠ), photosystemⅡ(PSⅡ) 등 여러 가지가 존재 한다. 이번 실험에서는 plant의 leaf에 있는 이러한 protein을 분리하여 어떤 protein이 plant의 leaf에 어느 정도 존재하는지 알아보는 실험을 실시하였다. 실험에 사용한 plant는 rice와 arabidopsis이며, chlorophyll을 채취하는 실습에는 spinach를 사용하였다. 실험에는 대표적인 protein 분리방법 중 하나인 native-PAGE를 이용하여 분석하였으며 native-PAGE와 SDS-PAGE의 차이점에 대해서도 이해한다. 이 실험 결과 다른 plant와 마찬가지로 rice와 arabidopsis의 leaf에도 RC-LHC(Reaction Center-Light Harvesting Center), RC-core(Reaction Center-core), LHCⅡ trimer, SC(small complex), FP(free pigment) 들이 관찰 되었고, rice 와 arabidopsis의 pigment-protein의 구성에는 약간의 차이가 있음을 알 수 있었다.참고 자료
