1. Gram stainingBrief : The preliminary classification of a bacterium as gram positive or gram negative is an essential step in both diagnostic microbiology and clinical medicine.
The Gram stain is generally the first procedure performed in the identification of a bacterium and the results of this differential stain often determine the subsequent identification procedures.
Often Gram stain yields equivocal results particularly with gram positive anaerobic bacteria and anaerobic gram negative bacteria.
Protocols :
1. Prepare the slide glass and blow away the oils with the flame
2. Drop the bacterial broth on the glass
3. Heat fixation with the alcohol lamp flame
4. Apply on Primary staining agent(Crystal violet), 1 min
5. Wash the CV and drop the iodine agent, 1 min
6. Wash with the alcohol
7. Do the Counter staining(Safranin), 1 min
8. Wash with water and drop the mineral oil
9. Observe the results of staining with microscope
Result :
Our sample has 2 kinds of bacteria. Purple coccus, and pink bacillcus
