1. Principles
1) Distribution ratio (K) or (Partition coefficient)
An analyte is in equilibrium between the two phases;

where [C S ] is the concentration of a solute in a unit volume of the
stationary phase, and [C M] is the concentration of a solute in a unit volume
of the mobile phase.
2. Classification
Cf) Affinity chromatography is a chromatographic method of separating
biochemical mixtures, based on a highly specific biologic inter action such as
that between antigen and antibody, enzyme and substrate, or receptor and
ligand.
3. Theoretical aspect of Chromatography
1) Plate theory
: the column is divided into a number of zones called theoretical plates and
the column assumed to be ‘static‘ system in equilibrium (assume s that
equilibration is infinitely fast).
A. A Theoretical plate: an ideal section of the column where the
concentration of the solute reaches equilibrium between the mob ile
phase and the stationary phase (ideal column: infinite number).
B. The number of theoretical plates in a column ( N) and the plate height;
the Height Equivalent to a Theoretical Plate (HETP) serve as a way of
measuring column efficiency . If the length of the column is L, then the
HETP is
HETP = L / N
N : (the more plates, the better), HETP: (the smaller, the better).
