Introduction
- Selenium is and essential trace element for animals and humans.
- 80% of body selenium was found to be protein bound.
- selnium-containing proteins or subunits in tissues of mammals have been reported, glutathione peroxidase (GPx), selenoprotein P, albumin, type-1 iodothyronine 5`-deiodinase.
- Selenium distribution among albumin, selenoprotein P and plasma-GPx, in human plasma or human serum was investingated by high-performance liquid chromatography (HPLC) coupled with a fluorescence detector (FLD)
- 21±1 kDa protein should be plasma glutathione peroxidase (p-Gpx), Se content was about 21.1-24.3%.

- Sample Collection and Pretreatment
: Human blood sample_ serum, plasma
- Selenium Determination
Fraction digestion
① CEM microwave digestion system (Model MDS-2000) with temp 200℃
② pooled fraction : mixed with 10vol of HNO3 in Teflon digestion vessel
1) Selenate in the digested solution : reduced to selenite in water bath 100℃
with equal volume HCl
2) add 0.5ml of 0.1M EDTA, 1ml 2% HONH2HCl (Hydroxylammonium chloride) pH1.5~2.0
