Procedure
[Team5] MRS Agar culture media
1. Weigh certain amount of each materials and put all materials into the 1ℓ Erlenmeyer flask.
⇒ Materials : Lactobacilli MRS broth 27.5g, Agar 10g
2. Put 500㎖ of primary distilled water into the Erlenmeyer flask.
3. Close the inlet of the Erlenmeyer flask with aluminium foil
4. Put the Erlenmeyer flask into the auto clave and sterilize it. (at 121.1℃ during 20 minutes.)
5. After sterilization, cool down the flask until the temperature of the Erlenmeyer flask is approximately 50 ~ 60 ℃.
⇒ The medium hardens in less than 44℃, so do not cool down too much time.
6. Pour a liquid medium which cooled enough onto a Petri dish by 30㎖.
⇒ This process should proceed in the clean bench.
7. After about 10 ~ 20 minutes, when the medium harden, close the Petri dish and turn over the Petri dish.
8. Keep and solidify the medium in the clean bench during one day, and seal each Petri dish with parafilm.
9. Storage the media in the refrigerator. (at 4℃)
