세계적으로 colorectal cancer (CRC) 는 여성에게는 the second most common cancer, 남성에게는 the third most common cancer 로 알려져 있으며 국내의 경우 여성의 the third most common cancer, 남성의 the second most common cancer로 조사 되어 있다.
1) Mechanisms of carcinogenesis
CRCs 는 chromosomal instability (CIN), CpG island methylator phenotype (CIMP), 그리고 microsatellite instability (MSI) 의 different mechanisms 의 하나 혹은 조합에 의해 발생한다.
2) Chromosomal instability (CIN)
Chromosomal instability 는 sporadic CRCs 의 65%-70%와 관련이 있다.
classical CIN pathway는adenomatous polyposis coli (APC) 의 mutations 의 획득으로 시작하여 oncogene KRAS 의 mutational activation 과 tumor suppressor gene, TP53. 의 inactivation 이 이어서 나타난다. Aneuploidy and loss of heterozygosity (LOH) 가 CIN 의 major players 이다.
