1주차Genetics: 유전(heredity)를 연구하는 학문 (시간에 따른 유전)
Genomics: 유전체, 개체의 유전자를 전체적으로 다루는 학문 (유전체의 공간)
Ex) 암을 유발하는 특정 유전인자가 어떻게 유전되는지 통계학적으로 접근(Canccer genetics), 한 암 환자의 유전체에서 발생한 변이들의 위치 및 pathway network 파악 (Cancer genomics)
- 후성유전학(Epigenetics, Epigenomics)
: DNA sequence의 변화 외의 것들을 연구
Ex) Chromatin의 구조에 따른 유전 및 발현 양상, methylation 등 DNA의 화학적 변화, Histone tail과 전사인자(transcription factor)의 결합
→ 이러한 것들이 신체 부위별 유전자 발현이 다르게 나타나도록 유도
DNA는 유기화합물로 다양한 생화학적 반응이 가능
환경적인 요인(영양 섭취 등) 또한 유전 발현과 연관이 깊음
Ex) 영양실조 상태에서는 DNA methylation을 통해 발현을 조절하기 위한 재료가 부족, 2차 세계대전 이후 네덜란드 대기근으로 인해 당시 출생한 아이들의 유전병 발병확률이 높아짐
Chapter 1 - What is Epigenomic?
1.1 Chromatin and gene expression
Chromatin: genomic DNA으로 nuclear protein(histone)이 감싸져 응축된 형태
→ super coiling되어 chromosome 형성(22쌍의 상염색체 + 1쌍의 성염색체)
Heterochromatin(closed): 전사 인자가 결합할 수 없는 상태, metaphase of mitosis(세포분열의 중기)에 일시적으로 가장 고밀도로 응축됨
Euchromatin(opened): 유전자 발현이 일어날 수 있는 상태
