Ⅰ. 입문 (Introduction)
1. lac-Operon
1) lac-Operon 정의
Operon이란 연관된 유전자들이 통합적으로 조절될 수 있도록 하나의 전사 단위로 묶여 있는 것을 말하며, lac-Operon 은 lactose를 glucose+galactose로 분해할 때에 관여하는 operon이다.
2) lac-Operon 구성
① Promoter : RNA Polymerase(RNAP)가 결합하는 부위
② Operator : repressor가 결합하는 부위
③ Structure genes : a) lacZ - β-galactosidase 발현
b) lacY - lactose permase 생성 (lactose를 박테리아 내부로 운반)
c) lacA - thiogalactoside transacetylase 생성
2. Positive & negative Regulation
1) Negative Regulation
① Lactose가 없을 때
Regulatory gene에서 repressor 항상 발현 ⇒ operator에 결합하여 RNAP의 결합 방해
② Lactose가 있을 때
Lactose가 isomer인 allolactose로 일부 변환
⇒ allolactose가 repressor와 결합
⇒ repressor의 단백질 3차 구조가 변화되어 operator에서 떨어짐
⇒ RNAP가 promoter에 결합 하여 lac transcription 시작
2) Positive Regulation
① Glucose가 적을 때
cAMP (Hunger Signal) ↑
⇒ cAMP가 CRP(Catabolite Regulation Protein)와 결합
⇒ CRP-cAMP complex가 promoter 옆에 결합
⇒ RNAP의 promoter 결합 촉진
② Glucose가 많을 때
cAMP ↓ ⇒ CRP가 inactive ⇒ lactose가 있더라도 낮은 수준의 lac transcription
3. Diauxie
Diauxie란 두 가지 당의 혼합물에서 microorganism이 더 유용한 당을 먼저 다 사용한 후에 다른 당을 쓰는 것으로 인해 2단생장곡선을 보이는 것을 말한다. glucose와 galactose의 경우 glucose가 첫 번째로 다 사용된 후에 galactose가 쓰인다.
