1. Introduction
⑴실험 제목 : Identification of serum glycoproteins by SDS-PAGE and Western blotting
- SDS-PAGE gel preparation and running the samples & Western blotting
⑵실험 목적 : SDS-PAGE의 구동원리를 알고 직접 gel을 만들어서 electrophoresis를 한 후 Coomassie Blue staining과 Western blotting을 사용하여 probe하고자 하는 protein을 확인한다.
⑶실험 상의 개요
①일단 우리가 실험에 쓰고자 하는 protein은 다음과 같이 얻어졌다.

=>SRP30c-flag를 coding하는 sequence를 가진 vector를 transfection한 cell과, vector만을 넣어준 cell(앞의 cell과 동일)에서 각각 protein들을 얻음.
②이렇게 얻은 positive, negative control들을 2개의 pair로 나누어 SDS-PAGE를 걸어서 separation시킨 후, 한쪽은 Coomassie Blue staining을 하고, 다른 한쪽은 Western blotting을 한다. Western blotting에서의 개요는 다음과 같다.

=>primary antibody로 SRP30c의 flag를 detection하며, 이는 다시 antibody에 conjugated된 HRP이 ECL solution과 만나서 빛을 내는데, 이를 film에 감광시켜 SRP30c의 위치를 알아낸다.

①Glass plates, spacers, comb, electro-gel running kit
②30% acrylamide, bis-acrylamide stock sol. (29:1) (store at 4℃)
1:1 gel buffer( 0.75M Tris, 0.2% SDS, 0.1% TEMED)
