1. 실험제목 : Magnetically switchable glucose sensor using enzymes immobilized in mesocellular carbon foam
2. 실험목적 : Magnetically switchable glucose sensor에 관련된 기본 법칙들을 이해한다.
3. Materials & methods
⑴washing of Mag-MCF-C
①Mag-MCF-C(magnetic mesocellular foam-carbon) powder를 DDW에 용해시킨다.
②30분 동안 shaking하고 13000rpm에서 10분 동안 centrifugation한다.
③ ②의 과정을 2번 더 반복한다.
④supernatant를 제거하고 50mM sodium phosphate buffer(pH 7.2)를 더한다.
⑵Enzyme loading in Mag-MCF-C
①glucose oxidase solution(0.6mg/ml, 500㎕)을 준비한다.
②Mag-MCF-C 3mg을 resuspend한다(sonication & pipetting)
③13000rpm, 4℃에서 10분간 원심분리하거나 magnet collection을 10분간 실행한다.
④supernatant를 제거하고 enzyme solution을 가한다.
⑤250rpm, 16℃에서 20분간 pipetting & shaking하여준다.
⑥13000rpm, 4℃에서 10분간 원심분리하거나 magnet collection을 10분간 실행한다.
⑦supernatant를 제거하고 50mM sodium phosphate buffer(pH 7.2) 500㎕를 가한다.
⑧ ⑤~⑦의 과정을 2번 더 반복한다.
⑧supernatant를 제거한 후 50mM sodium phosphate buffer(pH 7.2) 150㎕를 가한다.
