what is HPLC?
The fact that seven herbicides can be separated in 6 min is taken for granted nowadays, reflection the rapid progress made in liquid chromatography since about 1970. Up to that time, only gas chromatography could provide this performance level. Separations in columns using liquid chromatographic techniques often took several hours. Thin-layer chromatography reduced the minimum analysis time to less than 1 h, but direct results are either qualitative(position of spot) or, at best, semi-quantitative(evaluation takes a good deal of time and money and does not provide a fully satisfactory solution to the problem anyway.
Liquid chromatography is also the fastest way of determining the presence of caffeine in drinks. Ready-made coffee, tea or Coca-Cola is simply injected into a suitable column and a quantitative signal is obtained after 5 min.
these examples show that high-performance liquid chromatography(HPLC) is very efficient, i.e. it gives excellent separations in a short time. The `inventors` of modern chromatography, Martin and Synge, were aware as far back as 1941 that, in theory, a high pressure is essential for forcing the mobile phase through the column. As a result, the HPLC is sometimes referred to as high-pressure liquid chromatography.
