5. Principle
1) Agar
It serves to solidify the growth medium. Characteristics of agar used in culture medium.
①It is hard and has no effect on the spread of matter.
②Maintain melting and jell-phase at a certain temperature.
③It is transparent when it is solid or liquid, so that it can distinguish the shape of the colonies.
2) Culture medium
Order to cultivate the microorganisms or the tissues of plants and animals, the nutrients required for the cultures are the main components, and the substances for special purposes are mixed again. It supplies carbon, nitrogen source, inorganic salts, growth factors (vitamins), etc. as nutrients as well as water indispensable for survival and development except gaseous phase.
Culture medium are greatly divided according to type solid medium, liquid medium.
Solid medium are divided into four categories.
