Purpose:
The purpose of the experiment is to analyze the effect of restriction enzymes HindIII and EcoRI on the DNA molecules undergoing PCR and discuss further applications of the enzymes.

Circular DNA molecule has no ends as it forms a closed loop of DNA. The circular DNA in the nucleoid region of bacteria plasmid plays essential roles in terms of genetic variations and evolution. Bacteria such as E.coli transfer genetic material from one to another by conjugation of plasmids. When such circular DNA molecule is cut once with a restriction enzyme, it will no longer be enclosed in a loop and end up with a single, linear DNA molecule.
