1. 실험 이론 및 원리
가. 실험 요약
본 실험에서는 time-resolved fluorescence spectrometer를 이용하여 Cy3 fluorophore donor와 Cy5 fluorophore acceptor에 대해 Foster resonance energy transfer (FRET)에 의해 나타나는 fluorophore emission spectra를 측정하였다. 또한, 565㎚에서의 Time correlated single photon counting (TCSPC) 를 이용하여 fluorophore intensity decay curve의 측정을 통해 fluorophore lifetime을 계산하였다. 또한 이를 이용하여 FRET efficiency를 계산하고, 이론적인 계산값과 비교 및 분석하였다.
나. Fluorescence
Fluorescence는 molecule이 에너지를 흡수하여 들든 상태에서 존재하다가 바닥 상태로 내려오며 그 에너지 차이 만큼의 빛을 자발적으로 방출하면서 나타난다. Figure 12에서 나타낸 이러한 과정에는 phosphorescence도 존재하는데, phosphorescence와는 달리, fluorescence는 수 ns 이내 에 방출되기 때문에 빠른 시간 내에 일어난다는 점에서 차이가 있다. Fluorescence의 decay 과정에서는 radiative pathway와 nonradiative pathway가 존재하고, 이 과정에 따라서 emission wavelength가 다르게 나타난다. Fluorescence 측정 방식에는 steady-state measurement, time-resolved measurement가 존재하는데 본 실험에서는 fluorescence가 decay되는 것에 걸리는 시간보다 더 짧은 시간 동안 pulse를 내보내는 방식인 time-resolved measurement를 이용하여 fluorescence decay를 측정하였다.
