Abstract
이번 실험은 C3H/He strain의 7.5일이 된 생쥐 embryo를 다른 생쥐의 정소에 이식하여 발생된 다양한 세포와 조직들로 이루어진 teratoma를 분리한 세포로부터 구축된 embryonic carcinoma cell인 P19 cell line의 pluripotency를 이용하여 3배엽으로 만든 뒤에 Retinoic acid를 이용하여 P19 cell를 뉴런으로 분화를 유도하였으며 이를 RA 처리하지 않은 negative control과 marker gene인 Pax6 발현 차이를 비교하였다. 처음 P19 세포를 petri dish에 배양하여 Embryoid body가 형성되도록 만들었으며 중간 기간에 media change를 하였다. 그 뒤에 이를 tissue culture dish에 배양하여 어느정도 분화를 시켰으며 8일차에 RNA extraction을 진행하였다. 순수한 RNA만을 추출한 뒤에 이를 RT-PCR로 cDNA를 합성하였으며 최종적으로 electrophoresis를 통해 분리하였다.
Introduction
정자와 난자가 결합해 수정란이 되고 하나의 세포로 시작되는 수정란은 cell division을 하여 여러 개의 세포로 이루어진 blastocyst가 된다. Blastocyst 안쪽에는 inner cell mass라 하는 세포 덩어리가 존재하는데, 이 세포 덩어리는 cell division을 거쳐 embryo를 형성하게 되고 embryo는 하나의 개체로 발생하게 된다. 이러한 과정에서 inner cell mass의 세포들이 뼈, 피부, 혈액, 간 등의 한 개체에 있는 모든 조직의 세포로 분화하게 되는 것이다. 이런 inner cell mass의 cell을 blastocyst로부터 분리해 특별한 환경에서 배양하면 더 이상의 division은 일어나지 않지만 분화할 수 있는 능력은 보유하고 있는 상태의 세포를 만들 수 있는데 이런 세포를 embryonic stem cell이라 한다. Embryonic stem cell은 모든 조직세포로 분화할 수 있는 능력을 가지고 있으며, 이론상 무한정 cell division을 할 수 있다.
