Introduction

- Transformation is the genetic alteration of a cell, uptake and expression of foreign genetic material (DNA or RNA).
- Competent cells are bacteria which can accept extra-chromosomal DNA or plasmids. Cells can be made competent in several ways.

Chemical methods
1. Hanahan method
2. Inoue method incubating bacterial cultures at 18℃
3. Calcium chloride method using complex cocktails of divalent cations

Physical method
1. Electroporation method Exposure to an electrical charge destabilizes the membranes of E. coli and induces the formation of transient membrane pores through which DNA molecules can pass.

v Calcium chloride method
The calcium chloride ions neutralize the repulsion between the negatively charged phospholipid heads of the cell membrane, and the negatively charged phosphate groups on the DNA.
