Insulin and insulin analogs
The highest concentration of endogenous insulin first passes through the liver, suppressing the liver`s production of glucose by gluconeogenesis.
The quantity of injected insulin required to adequately suppress hepatic gluconeogenesis and to drive glucose out of the blood and into insulin-dependent cells is greater than that produced physiologically.
Insulin and insulin analogs
Insulin lispro
Regular insulin has its onset in 30-60 min and peaks 2-3 h after subcutaneous administration. In contrast, insulin lispro has its onset of action within 15 minutes of injection and has its peak action within 30-90 minutes.
type 1 diabetes and GDM, postprandial glucose and Al C concentrations and hypoglycemia have been reported to be lower in women using lispro in comparison with those using regular insulin.
One rationale for the use of all insulins is that unless antibody-bound, the drug does not pass the placenta.
