<예비보고서>
1. 실험제목 : Isolation, subfractionation, and enzymatic analysis of beef heart mitochondria
2. 실험목적 : beef heart muscle에서 mitochondrial fraction을 분리하고 protein content에 대해서 분석하며, 각각을 submitochondrial particle로 fractionate하여 malate dehydrogenase & monoamine oxidase activity에 대해 분석한다.

3. Introduction & Content
⑴Mitochondria의 구조
①double membrane : smooth outer membrane(permeable), inner membrane(unique transport & protein, enzyme 존재)
②intermembrane space, matrix : 각각 다른 생물학적 기능을 나타냄

⑵mitochondria의 기능
=> aerobic (oxygen dependent) metabolism of the cell에 관여한다.
①cristae에 존재하는 효소들은 citric acid cycle, respiratory chain, and beta fatty acid oxidation에 필요한 효소들이다.
②독립적인 DNA를 가지고 있어 protein enzyme에 필요한 mRNA 합성이 가능하다.

⑶intact mitochondria의 분리
=>크기가 크기 때문에 분리하는데 유리하다. 주로 쓰는 방법은 differential centrifugation of homogenized tissue이다.
--->mitochondrial fraction(major compartments), lysosomes, cell fragments, nuclear fragments, microbodies(peroxisomes)로 분리되며, 다음과 같은 순서로 mitochondrial fraction을 준비한다.
