Ⅰ. Purpose
- Through TLC (Thin Layer Chromatography), the relationship between the isomeric structure and Rf can be found.
- A mixture of isomers can be separated through column chromatography (CC).
Ⅱ. Background
Chromatography: It is a physical method that separates the stationary phase and the mobile phase. Prepartive chromatography is to produce a pure substance through purification, and analytical chromatography is to measure the relative proportion of an analyte in a mixture.
1. Thin Layer Chromatography (TLC)
TLC is to analyze selective absorption by intermolecular interaction between silica gel and developing solvent. For example, if the material is polar, it is more bound to the silica gel and the solvent moves slowly. If it is non-polar, it moves more.
Usage example
A pure material will show only one dot on the TLC, and a non-pure material will show many spots (purifying). As the reaction proceeds, the reactant decreases and the product is generated to adjust the reaction conditions (monitoring-checking).
