1. Introduction
§ Acid-fast & endospore stain §- 항산성 염색법은 항산성(acid-fast)을 나타내는 Mycobacteria, Nocardia, Diphtheroids, Actinomycetes와 같은 미생물을 동정하기 위해 사용한다. 이들 세균들은 일반적으로 염색이 잘 되지 않으나 일단 염색된 후에는 산이나 알코올로 탈색해도 탈색이 잘 안되는 것이 특징이다. 염색과정에서는 가열을 해야 하고 염기성 색소인 carbol-fuchsin액을 사용한다. 항산성균이 이 염색액에 일단 염색된 후에는 탈색액에 내성이므로 염색된 미생물은 적색으로 나타난다.- 아포염색을 통해 영양 세포로부터 형성되는 아포의 크기, 형태 등을 염색하여 관찰한다.
2. principle
Nocardia
- Nocardia is a genus of weakly-staining Gram-positive, catalase-positive, rod-shaped bacteria. It forms partially acid-fast beaded branching filaments (acting as fungi, but being truly bacteria). It has total 85 species. Some species are non pathogenic; some species are pathogenic (nocardiosis). Nocardia are found worldwide in soil that is rich with organic matter. In addition, Nocardia are oral microflora found in healthy gingiva as well as periodontal pockets. Most Nocardia infections are acquired by inhalation of the bacteria or through traumatic introduction
- Nocardia grow slowly on non-selective culture media, and are strict aerobes with the ability to grow in a wide temperature range. Some species are partially acid fast (meaning that a less concentrated solution of sulfuric or hydrochloric acid should be used during the staining procedure) due to the presence of intermediate-length mycolic acids in their cell wall. Majority of strains possess the cord factor (trehalose 6-6` dimycolate) an important virulence factor.
