Explant preparation and sterilization
① Collect carrot seeds.
② The seeds are sterilize for 1min in 70% Ethanol.
③ Discard the ethanol.
④ Seeds are sterilize with 70% Clorox containing 2 drops of Tween 20 for 20 min with
gentle shaking (150 rpm).
⑤ Rinse three times with sterile distilled water in the clean bench.
⑥ Dry the seeds with filter paper.
⑦ Seeds are culture on medium for germination in the dark at 25°C for 1 week.
⑧ Use the hypocotyls of plantlet after 1 week.
Callus induction and Regeneration
① Slice the hypocotyls about 4~8 mm with a blade.
② Explants are culture on callus induction medium in the dark 25°C for 4 wks.
* Subculture the calli to fresh medium every 2wks.
③ After 4 wks, transfer calli to regeneration medium.
④ Incubate on MS in the 16/8hours light/dark.
⑤ As they develop, transfer vigorously leafing out plantlets to half-MS for root induction.
