1) lac-Operon의 구성
Operon 이란, Promoter, Operator, Structural Gene등에 의하여 조절이 되고 있는 서로 이웃한 유전자군을 말한다. lac operon의 기능은 lactose 대사에 관계된 것으로, E.coli가 배지에서 glucose가 고갈되었을 경우 다른 에너지원으로 lactose를 사용하기 위해 발현된다. 따라서 glucose가 풍부한 경우에는 조절유전자에 의해 작동을 하지 않다가 glucose가 고갈되었을 경우 작동하기 시작한다.

β-galactosidase (lacZ): beta-galactosidase를 coding한다. lactose를 glucose와 galactose로 가수분해한다.
lactose permease (lacY): glactoside permease를 coding한다.이는 외부에 존재하는 lactose를 cell 안으로 transport하는 transmembrane protein이다
3galactoside transacetylase (lacA): galactoside transacetylase를 coding한다.

lac repressor (lacI) : lactose가 존재하지 않을 때 operator DNA site에 결합하여 lac operon의 transcription을 repress하는 단백질
lac promoter (lacP): RNA polymerase가 lac operon에 binding하는 DNA site
lac operator (lacO): lac repressor가 binding하는 DNA site

2) positive/negative regulation
Negative regulation은 Lac-repressor에 의해 이루어진다. 이 Lac-repressor는 lactose가 없을 경우에 lac-operon에 속한 유전자의 발현을 억제한다.
